I just got the PO401 code on my Camry. I checked the EGR valve with a vacuum guage and it tested fine.
I used a Haynes manual procedure to check the modulator. They say to block the two ports that are together on the valve and blow air thru the single port with the engine off. You should get air flowing out the hole in the top of the modulator.
I then repeated the test with the engine at 2500 rpm and the book says the flow should be restricted. Mine wasn't at all. I also noticed when I pulled the modulator filter it had a black dot on the underside. Supposedly that could indicate the internal diaphram is blown. I think the test in the link above sounds more accurate.
I ordered the modulator and the VSV along with EGR valve gaskets. I didn't test the VSV but will replace it anyway while I take apart the EGR system for cleaning.

do you mean that the opening that is at bottom of modulator where hose is used to connect to egr valve?
or
i have to open the modulator to do this test
