Has anyone tried installing an S54 on a 5SFE? What are its drawbacks?

your thinking of the E53. (not very popular for some reason). the gear ratios are super wide and would only make a 5s powered car slower then it already is.
S51=Camry 5sfe
S53=Celica 5sfe
S54=MR2 5sfe(same as 51 but with the shifting forks relocated on the housing for easier connection from the front)
E53=Camry 3vzfe
E153=Camry SE 3vzfe
Celica 3sgte(need confirmation for 3s)
only the E153 can be fitted with the Quaiff or TRD LSD
Adam correct me if I'm wrong...
Both S51 and S53 were used on the gen2 Camry's. I have an S53 in mine, though i've seen a lot more with the S51.
I believe the 3S-GTE uses the E153 as well; very common with the turbo MR2's which use that combo from factory IIRC. Stronger internals i suppose.

You mean to say S51 and S54 have the same gear ratios?
