I believe youa re correct onthe location. I think there is a door type flap you can open to remove and install the filter. I seem to remeber hearing it should be done anywhere between 15-30k miles depending on the auto manufacturer. Of course with my Gen 3 I dont ahve to wrry about it since there isnt one. I did not know the Gen4 's had one either. I dont even think my Sienna has one and it is an 02 XLE loaded.
You should also be able to find info on replacing and the location in your owners manual.

hmm... i have some little chunks of foamish stuff coming out of my vents. they are foamish because they like.... fall apart hella easily. and when you press on the little pieces, they are like marshmellows haha. what is this? its like you can dissolve it in water or something. ist there something i need to replace? ><
My Gen4.5 does have it. You have to remove the carpeting on the drivers side (RHD) footwell. It is near the footrest. It's quite durable, 6 years old and I just vacuum it and reuse it.
the foam that you are seeing is an insulation that goes around the vents to help keep the air from leaking out around the vent. Over time the foam starts to disentigrate and will sometimes make its way out of the vents.

My gen 5 has it. They want you to replace it every 15K miles. I have 100K miles on my 04, so that was way to much money for me. I just take it out and clean it every oil change.
