How much will it cost?? I just came from Autozone and they told me I have Cylinder 1 and Cylinder 4 misfire. Can I still drive my car like this?

Cost will depends on what is causing the misfire.

You can drive the car...but I'd recommend against it. It will cost you in the long run.

start simple check your plugs and wires. If you have a bad plug or an old wire with a lot of resistance you can burn up your coil or coil packs.

If you have one bad wire, that will cause the 1 and 4 cylinders to misfire
To ckeck for bad insulation take a spray bottle and fill it with water.
Start the car and raise the hood and mist the driverside of cylinder head, then along the wires.
If you see sparks, then the wire insulation is bad.

If you do what Hajoca said and you see a little blue spark that could be causing the misfire. A misfire is when a particular cylinder is not firing or firing at the wrong time, many different things can cause it. Wires and plugs would be a good pace to start if they have not been done in a long time. I would not drive it as excess gas from the missfire can burn out your cat which = more money!

^It could be. Or, it could be something else. Good luck with that.

I was starting with the cheapest, least messy and easiest thing to do.

What year is this engine??

The firing of the spark plugs (timing) is all handled electronically with a crank position sensor, the advance or delay is handled by the PCM and the signal is sent to the coil pack to fire.

The next thing to do is take out the spark plugs and inspect those.

If they are fine, you ned to check for spark. Got to a dark/dim location, then open the main fuse compartment (next to battery) and pull put the EFI fuse. Then connect the spark plugs to the plug wires, and lay them on the valve cover.
have an assistant crank the engine while you are watching the plugs. you can stay 4ft back to do this. crank for 2 seconds, and you sould see bright sparking at the plugs.

If not, then you have narrowed it down to wires or coil packs.

Use ohm meter to check the wires from sparkplug end to the coil end. around 20kOhm if I remember is ok. Bend and flex the wires when you check to look for bad spots. Any higher resistance, then you will need new wires. the total resistance if wires 1+4 should be about same for wires 2+3.
