I have Clarion 6X9's in the front doors. as you guys know, there's a huge hole in each door covered by clear plastic. doors act as terrible boxes to begin with, but if I seal those big holes with dynamat or with thin fiberboard, will that improve midbass response?
I dont know what car you are talking about, but speakers do not really depend on the enclosure for the air movement. In your case, covering the holes might help, but it might not be too noticeable of a difference. Dynamat IS pretty expensive.
On the contrary, the enclosure is dependent for air movement. I would use sound dampener to cover as much of the inner and outer skin of the door as possible(behind the door panel). There wont be a huge difference, but it will help, a.to reduce the volume of the "enclosure" of the speaker aka the door, and b. reduce the vibrations of the door panel which in turn reduces the amount of energy lost.
deadening your door will increase the midbass. make sure to fill in the holes with something that's pretty solid, plexi, aluminum sheeting, etc. Sealing off the door as much as possible will help with your midbass output. If sealed and deadened properly, there will be a significant difference.

Look up the foam baffles that are made to go in the speaker holes. Those will help out a little bit, and if your sure they are clearing the window, soak em in fiberglass resin and make them rock solid. That should greatly improve your bass response.
