I wouldn't say its serious as long as you can shift all the gears, though it could move and block the shifter from going into park possibly so it might need to be removed. Its fairly easy to get to that area of the car.
Take a look here (DIY Radio and Subwoofer Install in 2009 Toyota Highlander). Look at the 8th picture where Luck123 removed the panel that holds the cigarette lighter and aux in jack. That will need to be removed. Then you can reach into the opening and grab the plastic center console and pull straight up. It will remove the whole plastic cover that is over the shifter and the 4 cup holders (everything in front of the center storage compartment). There is a single large plug that has all the wires for the buttons and cigarette lighter on that plastic cover, unplug that and you have easy access to the shifter components. It is fairly easy to disassemble the shifter components and get to the quarter if you just take a look at it and take your time. It was a few weeks ago I did this so I can't remember how to disassemble the shifter itself, but I do remember it was straightforward.

I would like to remove that cover as well. In the series luck123 posted he had one end open already. With nothing removed at which end (side) does one start? You need to insert something (screwdriver?) to start it. I take it the center storage compartment stays..

You start on the side next to the radio. The center storage compartment will stay in place, it will just be the cover that is removed. I used a screwdriver to get the first dash piece off (the one holding the cigarette lighter, aux in, etc...) Everything else was by hand

This seemed so simple.. Perhaps I am missing something as this is as far as I get. The back comes up easily enough but the front is tucked under the dash part. It seems like the thing "should" slide straight back at this point.. but does not I tried removing just the inside cover.. this is what I assumed the instructions were, the edges lift up but there is something very solid holding the center, the electrical connections to the lighter I am guessing.

Your going at this the wrong direction. That little plastic piece you have on the driver seat does not need to be removed, nor do the circle trim pieces around the cup holders.. The part in the red box (that whole peice) needs to be removed first (look in luck123's thread for a how to) then the whole panel comes straight up just like the back did. There is a large electronic connector (for the 12V outlet, downhill assist button, and the other button there, I think snow) under the plastic panel at the edge under the green square that will have to be disconnected, then your good to go. So remove the red, disconect the green and your set. Sorry if I was confusing before, I wasn't really sure what you were asking in your last post

Thanks Patric, I will give it another shot tomorrow. Just to be clear the part enclosed in your red box comes out (the part that has a little door over it), the side bits do not? I think there may be some differences since luck123 did his piece..

The red square and sides are connected. Look here (DIY Radio and Subwoofer Install in 2009 Toyota Highlander) at pic 8 9 and 10 it shows the part that has to be removed. it is that whole section.
EDIT: just way ur edit, I still think the sides are connected. The different car trims levels (standard, se, lim) have different centers, but the sides should be the same
