When I cross a particular overpass near my house, in one direction, I hear a sound that is sort of a cross between an animal being mistreated/squeaky bike/birds chirping. It appears to come from the front passenger door speaker area. It happens even if the radio is off.
The first time it happened, I inspected the entire interior area literally looking for a bird or some animal that had gotten inside the car.
Given that it usually only happens when I cross this particular overpass, I suspect there may be some electrical interference from something nearby, but it's odd that it happens even if the radio is completely turned off.
Any suggestions?

trucks with premium sound systems often have physically separate amplifiers distributed throughout the vehicle--I've never looked at 2nd Gen HL's to see hoe they do it. It's possible that these amps remain on or are in a standby mode with the head unit off. Some amplifiers are designed to "wake up" or come out of standby as soon as they see signal/audio on their inputs. I've seen in other applications where RF interference has triggered this.

No. No. No. Forget about all this science stuff your going to hear in response to the problem you have posted here. As a certified "Run Away, (not merely wander from the flock), at the age of 9, Catholic", I can tell you what you really must do in this kind of situation. You need to stop by any Catholic church and, to the left and right of the main doorway, you will find some water, (this is not a drinking fountain), this is Holy Water! Bring a small container and dip it in, then take it out to you car and toss it on it as you say "Demon's Be Gone", in clear, firm voice. (If you have a priest or nun do this it could cost you a lot of money). If the sound is gone the next time you go over that overpass, your in the clear. If it's still there then start considering all the sort of tech, science ideas you're going to get. Good Luck!

anyway,
the JBL amp is on the entire time the car is on (even when the head-unit is off), so I don't think its turn on noise.
are you sure its coming from the speaker, it could be some kind of equipment near that overpass causing electronic interference in that speaker (from a not very well shielded connection or wire maybe...), or the road is maybe rough or bumpy there and something is squeaking in the door/ that area

Possibly you are getting a reflected noise that normally you do not hear. Perhaps the road surface in this area is also contributing to a different noise. Does it vary with speed? Try stepping on the brake lightly while you hear the noise to see if it changes.
I have the base model (without JBL), but there may be an amp anyways.
It's not background noise, because when it happens, its very loud. It lasts for a few seconds, and then goes away. The overpass is right next to a Toyota dealership, so if it keeps up I may demonstrate it to them.

Hmmmm. This is sounding suspiciously like a cry for help from your HL. You may have a very homesick Highlander. This is a very unusual condition, have you mistreated your HL much? They do thrive on TLC
Cell tower adjacent to the bridge? Cell transceivers mounted to the bridge structure? Emergency service transceivers mounted on or near bridge? Long-haul HV power lines overhead? Anything like this could induce noise bursts in live amplifiers.
