I'm confused about 2 things in the original post:
(1) Are you saying the truck cap doesn't lock at all? Or that it locks, but you just don't think that's secure enough? If the former, you might be able to add a lock (e.g., replace the hatch handle with a locking one). If the latter, OK...
(2) Why did you add the bit about being accessible from the cab? My experience is that by the time I twist and contort and futz with the slider windows, I could have just as easily walked around back... Is this a critical requirement?
Problem with bed chests / trunks is that the manufacturers can't predict the inside geometry of your cap (to clear their lid), so they figure it's a brain-dead notion to offer chests to go under a cap. If you're willing to drill the bed, I'd suggest finding a chest (e.g., a heavy-duty poly / plastic / metal one with a removable locking lid) and bolt the sucker in.
There's a very expensive product called BedVault (??? - something like that...) which provides customized (and even lockable / secure) storage drawers on the bed floor, but I suspect that's overkill in this case...
Personally, when I carry a laptop, it travels in a Pelican case inside the cab - period...
