Unable to remove spare tire

I am picking up my new Prius tomorrow and selling my beloved 2003 Corolla in the evening. When cleaning out the trunk we discovered some stuff of mine is stuck under the spare tire. We have to remove the tire to get it out but cannot figure out how to get the tire out. There looks like there is a screw thing we can turn, but it won't budge. Do we need a tool? or are we just weak?

worse come worse, if the item stuck in there is not that important, go with the car as gift?

Would letting some air out of the tire relieve any tension and help to turn the wing nut? Perhaps some lite tapping with a hammer...

put some hot water to dissolve the material you spill there, unless you had spilled epoxy there, for which u have to cut the tire.
