I'd have to say that the room in the second row (paired with the fact that you can change the center seat out quite easily) is something I like. Being in Minnesota I'm sure I'll love the heated wiper area once the snow starts to fly, and already the heated mirrors have come in hand on a frosty morning or two. The rear glass hatch is certainly nice (coming from an Explorer which had that as well, you get quite accustomed to just opening that instead of the entire rear door.)
The Nav, and associated things it brings, like maintenance reminders, is nice, but I also like the 'mini' screen that provides useful information, like "You left the moonroof cracked open, silly" when you turn off the car.
The one thing that has suprised me how quickly I've adapted to is the Smartkey. Call me goofy, but I just love that I can just walk up to the Highlander, grab the handle, and it's unlocking as I get in. I rarely use the fob/remote functions anymore, I just leave the thing in my pocket. The simple pleasure, I guess!
