I'm referring to the one nearest to the fender, not near the front grill. Any tricks to changing this one? Does anything need to be removed?

Thanks.

Battery must be removed for access. Remove Negative terminal first and replace it last.

Plastic cover above radiator must be removed to access front battery hold down bolt.
Factory cover push-in retainers become brittle over time and some or all can be expected to break during removal. Not a bad idea to buy new ones in advance at Lowes, Home Depot, or Amazon.

Refer to the drawing for location of the turn signal bulb. Twist a quarter-turn and withdraw.

Thanks. I'm beginning to wonder if this is a light or reflector. I thought I saw one side lit or flashing and the other side out but I could be wrong. The area I'm questioning is the amber, shaded area of the lens, next to the text "Clearance Light Bulb" in the graphic you provided. I can't determine if that is indeed the "clearance" (?) bulb or if that bulb is in another area of the lens.

That is the front "marker" light, which makes your vehicle more visible to others approaching from the sides. It is a separate bulb and is on whenever the tail lamps are on. On my Avalon, it is independent of the turn signal and does not flash..

When I look with a mirror and flashlight on the right side, where the battery does not obstruct the view, it is clear that bulb it not accessible without first removing the front bumper cover, and perhaps the entire headlight assembly. The headlight assembly is easily removed once the bumper cover is out of the way.

Instructions for removing the bumper cover can be found here:
