Our 2020 RAV4 has the same system and periodically prompts for a software update.You can't update the Entune firmware over the air. That "Check for Updates" button assumes you have inserted a USB drive that has an update file on it.
Here's the site where you can download such files and do that update if you really want to.

When I select Update Now and it downloads the update over the air and installs in a few seconds.
When the Highlander prompts for a software update the update fails.
