Timing Chain. That's one of the reasons I bought my 08. Possibly a coolant flush. Diff oil, check tranny fluid...I would have all the fluids checked. Cabin air filter.....maybe someone else can chime in with more important things.

coolant at 100k (50k intervals after this point on), standard oil changes/rotate. Do spark plugs at 125k. Everything is as fluid degrades.

 

Thanks for the input everyone! I was wondering about the spark plugs as well. I remember on my '06 Highlander I replaced the plugs and wires at 100K.

Haven't checked or looked but does the '12 also have wires?

Its coil on plug, no spark plug wires...or at least the kind your thinking of.
