Take a look at the Common Problems & Solutions sticky and click on the Steering Knock from the Intermediate Steering Shaft link, I think that'll answer your question.

 

It sounds like it's from the electric column assembly. Usually during turns and going over bumps. The vehicle has less than 400 miles on it. Wife isn't happy. Anyone else experience it too?

I can't find any service bulletins on it. The RX350 2010-11 also have this issue and there is service bulletin in regards to fixing this problem with a new steering column assembly. Almost similar design to the Highlander except the RX350 comes with electric tilt/telescopic.

Are you getting this "bump" jolting feeling throgh the steering column while driving straight over bumps also? I started a thread about this a few weeks ago....
